The principal objective of this Core is to provide PKD investigators with state-of-the-art proteomics and mass spectrometry. Specifically, the Core will provide access to these technologies and relevant education, either through on-site seminars and consultations or web-cast tutorials as well as hands-on-support for optimal utilization of the Core services. The Core staff can perform 2D-gel electrophoresis of samples and then visualize the gel spot pattern. The gel image(s) can be analyzed statistically with either NonLinear Dynamics Progenesis, or if labeled with Cy-dyes, with DeCyder. Investigators will be sent gel images and the statistical analysis to allow them to select the spots to be cut out of the gel and subjected to in-gel digestion by protease(s). These will be analyzed by MALDI-TOF peptide mass fingerprinting and the results analyzed with MASCOT to putatively identify the protein. Verification of the protein identity can be carried out by peptide sequencing by tandem mass spectrometry following nanoLCI-ESI Quantitative. Additionally, an important part of the support will be the archival of ongoing and completed experiments in recessive PKD into an existing proteomics database established through the UAB Proteomics/Mass Spectrometry Shared Facility through which PKD investigators can monitor progress of their sample analysis, but also be informed about proteomics data from other research efforts.

Planed services (beyond 2006):
Later in 2006, PKD Core Center Investigators will have access to MALDI Tof-Tof for more rapid tandem mass spectrometry analysis of peptides.
